Must be your Door Lock Actuator / Locking
Check this article it seems like the problem you are having.
Replacing the door lock actuator is necessary when the lock stops properly responding to lock and unlock commands from the remote or dash mounted toggle switch. The same procedure is followed for either door; however the driver side actuator does take a bit more time to install as it has the key lock linkage to also get back in place.

It looks like only fuse F37 is associated with the door lock: http://wds.spaghetticoder.org/en/svg/sp/SP0000017983.svg?m=r50
When you say you have to lock it manually, do you mean mechanically? Does the center console toggle switch work?
I haven't had any issues with the windows on my MINI, but I've seen lots of people posting about problems. Here's something to get you started: http://www.motoringalliance.com/forums/pittstopmini-club/1053-drivers-window.html-

I have to manually lock the doors. push the door button down or use the key in the lock. The central toggle switch will unlock but not lock the doors as well.

:cornut: HMMMMMMMMMMMM.................water/corrosion on electrical terminals somewhere along the line? That does things almost as strange as a battery going out.

The door lock actuator has been replaced. all fuses appear good. I am concerned it might be the Body Control module.
